Description
Lenalidomide is an immunomodulatory drug used in the treatment of multiple myeloma and other hematologic malignancies. It functions by binding to the CRBN E3 ubiquitin ligase complex, altering its substrate specificity to degrade key transcription factors (IKZF1, IKZF3). In myeloma, clonal evolution under lenalidomide selective pressure produces resistant subclones, and treatment cessation can shift the competitive landscape between sensitive and resistant clones.
