Description
Cisplatin is a platinum-based chemotherapeutic agent that forms intrastrand and interstrand DNA crosslinks, blocking DNA replication and transcription. It is used in the treatment of testicular, ovarian, bladder, head and neck, lung, and other cancers. Cisplatin-induced DNA damage produces a characteristic mutational signature detectable in tumor genomes. Resistance mechanisms include reduced drug uptake, increased DNA repair (NER, HR), and increased tolerance to DNA damage.
